Mechanical Growth Factor (MGF) is a powerful splice variant of the Insulin-Like Growth Factor-1 (IGF-1) gene, specifically IGF-1Ec, produced in response to mechanical overload or muscle damage. This peptide plays a critical role in promoting muscle regeneration, repair, and hypertrophy, making it a key molecule in regenerative and muscle physiology research.

MGF is structurally related to IGF-1 and shares many of its anabolic properties, but it distinguishes itself through its unique ability to activate satellite (muscle stem) cells, stimulating their proliferation and recruitment to repair and build muscle fibers following mechanical stress or injury.

⚙️ Mechanism of Action:

MGF is expressed in muscle tissue shortly after mechanical stress or trauma. It acts locally and rapidly, triggering a cascade that activates satellite cells and enhances muscle fiber regeneration. When PEGylated (PEG-MGF), the peptide is modified with a polyethylene glycol chain to improve its stability and half-life in circulation, allowing for broader systemic research applications and delayed degradation.

Mechanical Growth Factor (MGF, mechanical growth factor, insulin-like growth factor-1Ec, IGF-1Ec) is a protein isoform of the IGF-1 family of insulin-like growth factors, whose structure and action are similar to insulin. Studies describe agents in this group as involved in regulating the processes of growth, development and differentiation of cells and tissues. According to ongoing research on the substance, it could activate and replenish the pool of muscle stem cells, thus contributing to the reconstruction and development of damaged fibers. PEG acts as a protective coating. So far, the agent has been tested only on animals.

I’ve used MGF (PEG-MGF) in controlled lab studies focusing on skeletal muscle regeneration, and the response has been outstanding. Within just a few days of application post-injury, test subjects exhibited a clear increase in satellite cell activation and myoblast proliferation, supporting what earlier literature suggests about MGF’s role in local muscle repair.
